About this medicine

Xospata 40mg Tablet (Gilteritinib (40mg))is a prescription oncology medicine. Below is educational product information — follow your doctor's advice for dose and use.

Xospata 40mg Tablet is a prescription targeted therapy indicated for relapsed or refractory acute myeloid leukemia (AML) that works by inhibiting specific enzymes essential for cancer cell proliferation. Taken once daily at the same time with or without food, it should be used exactly as directed while attending all routine clinical follow-ups. Frequent adverse reactions include fatigue, gastrointestinal distress (nausea, diarrhea, constipation), musculoskeletal pain, and elevated liver enzymes, whereas emergency medical care is critical for symptoms like shortness of breath, chest pain, rapid weight gain, or sudden swelling. Because Xospata can alter cardiac rhythm, patients must disclose any history of heart issues and consult their physician before starting other medications or supplements. Finally, due to severe embryo-fetal toxicity risks, reliable contraception is mandatory during and after treatment, and breastfeeding must be avoided throughout therapy and for at least two months following the final dose.

Frequently Asked Questions

Take Xospata exactly as prescribed by your oncologist, typically as a 120 mg daily dose (three 40 mg tablets taken together once per day). Swallow the tablets whole with a glass of water, with or without food, at roughly the same time each day; do not break, crush, or chew them. If you miss a dose, take it as soon as possible on the same day, provided there are at least 12 hours before your next scheduled dose. If it is within 12 hours of your next dose, skip the missed dose and resume your regular schedule the following day—never take two doses within 12 hours to make up for a missed one.

Differentiation syndrome is a serious, potentially life-threatening reaction associated with rapid maturation of leukemic cells. It can develop anywhere from 1 day to several months after starting treatment. Seek immediate emergency medical care if you develop unexplained fever, sudden difficulty breathing or cough, rapid weight gain (over 4–5 kg in a week), swelling in the hands, legs, or feet, dizziness, or reduced urine output. This condition requires prompt intervention in a clinical setting with systemic corticosteroid therapy.

Because Xospata can impact heart rhythms and organ function, regular clinical surveillance is required: Electrocardiograms (ECGs): To monitor for QT interval prolongation; performed prior to starting therapy, on days 8 and 15 of cycle 1, and prior to starting subsequent treatment cycles. Blood Chemistries & Electrolytes: Frequent monitoring of potassium, magnesium, and calcium levels, as electrolyte imbalances raise the risk of cardiac arrhythmias. Liver Function & Pancreatic Enzymes: Routine blood tests measuring ALT, AST, bilirubin, amylase, and lipase to detect potential hepatotoxicity or pancreatitis.

Always consult your hematologist or pharmacist before starting any new medication, over-the-counter drug, or herbal product. Strong CYP3A4 inhibitors (e.g., ketoconazole, clarithromycin, itraconazole) or P-gp inhibitors can significantly increase Xospata levels in the blood, elevating toxicity risks. Conversely, strong CYP3A4 inducers (e.g., rifampin, St. John's wort, phenytoin) can decrease its effectiveness. Additionally, medications known to prolong the QT interval should be avoided or closely monitored when combined with Xospata

Xospata causes embryo-fetal toxicity and birth defects. Female patients of childbearing potential must undergo a pregnancy test before starting treatment and use an effective, non-hormonal or combined contraceptive method during therapy and for at least 6 months after the final dose. Male patients with female partners of reproductive potential must use effective contraception during treatment and for at least 4 months post-therapy. Breastfeeding is strictly contraindicated during treatment and for at least 2 months following the last dose.
